Tesis de Máster del año 1997 en eltema Biología - General, principios básicos, Nota: Apto, Universidad Autónoma de Madrid , Idioma: Español, Resumen: En la presente memoria de máster, se realiza un estudio de optimización y puesta a punto de diferentes técnicas inmunoquímicas para la detección y colocalización del péptido regulador neurotensina (NT) y su receptor (NTR) en células secretoras de la adenohipófisis de rata. Se estudia la expresión de NT y NTR en ratas macho y hembra a lo largo del desarrollo posnatal y en hembras adultas en, diferentes fases del ciclo estral, ovariectomizadas y ovariectomizadas con terapia hormonal de reemplazo. Se optimiza además, el protocolo experimental para el estudio de la coexistencia o colocalización de NT y NTR en los distintos tipos de células productoras de hormonas de la adenohipófisis. Por otra parte, se pone a punto la técnica de hibridación in situ no radiactiva para la detección del mARN del gen de la neurotensina/neuromedina N, y se discuten las particularidades, así como ventajas e inconvenientes de este tipo de técnicas en estudios de regulación neuroendocrina.In the present master's thesis, an optimization study of different immunochemical techniques for the detection and colocalization of the neurotensin regulatory peptide (NT) and its receptor (NTR) in rat adenohypophysis is carried out. The expression of NT and NTR in male and female rats is studied throughout the postnatal development and in adult females rats in, different stages of the estrous cycle, ovariectomized and ovariectomized with hormone replacement therapy. The experimental protocol for the study of the colocalization of NT and NTR in the different hormone-producing cells of the adenohypophysis is also optimized. On the other hand, the technique of non-radioactive in situ hybridization for the detection of the mRNA of the neurotensin / neuromedin N gene is being developed, and the particularities, as well as advantages and disadvantages of this type
